Spermidine has one of the strongest laboratory mechanisms in the longevity field and one of the thinnest human trial records. In yeast, flies, worms and mice it switches on autophagy, the cell process that recycles worn-out parts, and through it extends lifespan; that chemistry is well characterized in the laboratory. In people the evidence is smaller and softer.
Adults who eat more spermidine-rich food live modestly longer in observational studies, but people who eat those foods tend to eat well in general, so that is an association, not proof that spermidine caused it. The one larger memory trial found no effect on its main measure. For now the animal and dietary findings are promising and the confirming human trials have not been done, so the fair first step is food.

What It Is
Spermidine is a polyamine, a small molecule your own cells make, that gut bacteria produce, and that you take in from food. It was first found in semen, which is where the name comes from, but it is present throughout the body and turns up in almost everything you eat, most richly in wheat germ, whole grains, legumes, mushrooms, aged cheese and fermented soy foods such as natto. Your tissue levels of it tend to fall with age. The reason it draws attention is one well-studied action: it triggers autophagy, the process by which a cell breaks down and recycles its own damaged parts, and in laboratory organisms that action extends life.
What It Does
Spermidine's evidence splits into two halves that point in different directions. The laboratory work is strong and causal; the human work is thin and, where it has been tested directly, negative so far.
The laboratory half came first. In yeast, flies, worms and mice, spermidine switches on autophagy and lengthens life, and the same experiments show the life extension depends on that autophagy: block the recycling and the benefit disappears. In aged mice it also keeps the heart from stiffening. These results are consistent across species, and they are why spermidine moved from a biochemical curiosity to a longevity candidate.
The human half is smaller and softer, and it lines up in a clear order of confidence. Adults who eat more spermidine-rich food live modestly longer, but that is an observational pattern in people who tend to eat and live well in other ways, so it cannot show that spermidine caused it. When the compound was given as a supplement and tested directly, the one adequately sized trial found no effect on memory, its main measure. A smaller pilot had hinted at a memory gain earlier; the same small cohort supplied the short-term safety data, so it counts as one early signal, not two independent results.
The laboratory findings landed in 2009 and 2016, the dietary cohort in 2018, and the supplement trials after that. In order of confidence the findings run from a well-characterized mechanism, to consistent life extension in animals, to an observational dietary signal in people, to a null result the first time a supplement was tested for memory. Spermidine switches on autophagy, the process a cell uses to clear out and recycle its own worn-out parts, and that switch is what lets it extend life in simple laboratory organisms. When researchers block autophagy, the life-extending effect goes away.
Eisenberg et al. (2009, Nature Cell Biology) showed that spermidine inhibits histone acetyltransferase activity, triggers autophagy, and extends lifespan in yeast, flies, worms and human immune cells in culture. Genetic or pharmacological blockade of autophagy abolished the lifespan benefit, establishing autophagy as the mediating mechanism rather than a correlate. This is the well-characterized mechanistic basis for interest in spermidine.

No memory benefit from 0.9 mg/day spermidine over a year in 100 older adults
In the largest trial so far, older adults with memory complaints who took a daily wheat germ spermidine supplement for a year did no better on the main memory test than those taking a placebo.
The SmartAge trial (Schwarz et al. 2022, JAMA Network Open) randomized 100 adults aged 60-90 (49% female) to 0.9 mg/day spermidine from wheat germ or placebo for 12 months. The primary outcome, mnemonic discrimination on the Mnemonic Similarity Task, showed no between-group difference (-0.03; 95% CI -0.11 to 0.05; P = .47), and secondary outcomes were unchanged. Exploratory analyzes raised possible effects on verbal memory and inflammation that the authors said needed confirmation at higher doses.

A 30-person pilot hinted at a memory gain (Cohen's d 0.77) the larger trial did not confirm
A small three-month trial of 30 older adults hinted that a spermidine supplement helped memory a little. The study was too small to rely on and a larger follow-up did not confirm it.
Wirth et al. (2018, Cortex) ran a Phase IIa pilot in 30 cognitively intact older adults with subjective cognitive decline, using a spermidine-rich plant extract for three months. Mnemonic discrimination improved in the spermidine group with a medium effect size (Cohen's d about 0.77), though the confidence intervals were wide and reached zero. The authors framed it as a signal to be validated, which the larger SmartAge trial then failed to confirm on its primary endpoint.

Feeding spermidine helps yeast, flies, worms and mice live longer. This is an animal result and has not been shown to make people live longer.
Lifespan extension by spermidine was first reported in yeast, flies and worms (Eisenberg et al. 2009) and extended to mammals by Eisenberg et al. (2016, Nature Medicine), where oral spermidine increased survival in mice and was associated with enhanced autophagy and mitophagy. Many interventions extend rodent and invertebrate lifespan without ever replicating in humans.

Higher dietary spermidine tracks lower mortality, hazard ratio 0.76 per standard deviation (observational)
Adults who ate more spermidine-rich food died at lower rates over 20 years, and the gap between the highest and lowest intakes was about as large as being several years younger. This is an observed pattern, not proof the spermidine caused it.
Kiechl et al. (2018, American Journal of Clinical Nutrition) followed 829 participants in the Bruneck study (aged 45-84, 49.9% male) with repeated validated food-frequency questionnaires; 341 deaths occurred over 20 years. The age-, sex- and calorie-adjusted hazard ratio for all-cause death per 1-SD higher spermidine intake was 0.74 (95% CI 0.66-0.83), and 0.76 (0.67-0.86) after further adjustment for lifestyle and other dietary factors. The finding was independently reproduced in the SAPHIR cohort.

In aged mice, spermidine reduces heart stiffening and improves diastolic function
In old mice, spermidine kept the heart from stiffening and helped it relax properly between beats. This is an animal finding, not a demonstrated effect on human hearts.
Eisenberg et al. (2016, Nature Medicine) reported that oral spermidine in aged mice reduced left-ventricular hypertrophy and myocardial stiffness and improved diastolic function, with parallel improvements in mitochondrial function and autophagy. The paper also included human observational data linking higher dietary spermidine to lower blood pressure and cardiovascular events, which is hypothesis-generating rather than confirmatory.

1Inside the cell
Spermidine blocks an enzyme that adds acetyl tags to proteins, and through that and related routes it switches on autophagy, the process a cell uses to break down and recycle its own worn-out proteins and organelles. Fasting and calorie restriction switch on the same process, which is why spermidine is described as reproducing part of what they do.
2What autophagy clears
Worn-out mitochondria, misfolded proteins and other debris build up inside cells as the body ages. Autophagy is one of the systems that breaks them down and reuses the parts. Turning it up is the mechanism behind every laboratory result on this page.
3From cell to person
A compound can switch on a good process in a dish and in a mouse and still not change how long or how well a person lives, because human aging is slow and has many causes. That gap is why the mechanism is settled while the human benefit is not.
The spermidine supplements tested in people so far deliver around 1 mg a day, while an ordinary mixed diet supplies several times that from food, so the trials may have added little on top of what participants already ate. The autophagy process sits in the science library, alongside the wider picture of biological aging, and the food-first case runs through whole foods.

The mortality signal comes from food, and food supplies more spermidine than the trial supplements did, so the fair first answer is to eat the foods that carry it. The supplement rungs are for people set on trying the isolate, and belong in a conversation with a clinician.
Wheat germ is the richest common source, and whole grains, legumes, peas, mushrooms, aged cheese and fermented soy foods such as natto all contribute. This is the food pattern behind the observational link to lower mortality, and it overlaps almost entirely with a plain whole-food diet worth eating for many other reasons.
If you want to raise intake deliberately, a tablespoon or two of plain wheat germ stirred into yogurt, oatmeal or a smoothie is the cheapest concentrated food source there is. It keeps in the fridge once opened and delivers spermidine alongside fiber, B vitamins and vitamin E.
The supplements studied in people are wheat germ extracts standardized for spermidine, at roughly 1 to 6 mg a day. The one larger trial of such a product found no effect on its main memory measure, so it delivers the compound without an established benefit. Check for an independent testing mark, since supplements are loosely regulated.
One point to settle before spending anything: an ordinary mixed diet supplies several milligrams of spermidine a day, more than the roughly 1 mg in the trial supplements. That is part of why food-first makes sense here, and part of why the low-dose trials may have added little on top of what people already ate.
Taking spermidine in the hope of slowing your own aging is an experiment on yourself with an effect no human trial has measured. If you have a particular reason in mind, weigh it with the clinician who knows your history, and keep it alongside the habits with stronger human evidence such as exercise, sleep and diet.

The Chinese Medicine View
Spermidine is a modern isolate, named and characterized by chemists long after the classical Chinese pharmacopoeia was fixed, so it has no channel, temperature or flavor in the tradition, and assigning one would be an invention. What the tradition does have is a long way of reading the foods spermidine concentrates in, and that framing is laid over the modern compound as interpretation, held apart from the laboratory and trial data above.
The foods richest in spermidine are whole grains, legumes, mushrooms and fermented soy such as natto, and the tradition reads these mostly as strengthening to the Spleen and the middle burner, the digestive center it treats as the source of acquired vitality. Fermented and aged foods, the aged cheeses and the fermented soy, are read as warming and as helping the middle move, which suits someone whose digestion runs cold and sluggish. None of this maps onto autophagy or polyamine chemistry in any literal way, and reading it as if the tradition anticipated the modern finding would be a mistake.
Where the two views touch without merging is on who a food suits. The same fermented and rich foods that the tradition uses to warm and move a cold, damp middle burner are the ones it treats with more caution in someone already damp or phlegm-laden, where heavy, rich food is understood to feed the accumulation. That instinct, that the right food and the right amount depend on the constitution in front of you, sits comfortably beside a modern reading in which the benefit of a dietary pattern depends on the whole diet and the whole person, and it is offered as a way of thinking about the molecule.

Weigh who ran the trials
Several of the human supplement studies were designed or funded with involvement from a company that sells spermidine products, and some authors hold related patents or equity. That does not make the results wrong, and the studies disclosed it, but it is a reason to read the supplement claims with the source in mind and to lean on the independent dietary and mechanistic work.
The usual source is wheat germ, which contains gluten
Most spermidine supplements are extracted from wheat germ, and wheat germ contains gluten. If you have celiac disease or a wheat allergy, a wheat germ product is not for you; look for a certified gluten-free source or get spermidine from tolerated foods instead. This is easy to miss because spermidine itself is marketed as a pure compound.
Pregnancy, breastfeeding, active cancer, and children: check first
Supplemental spermidine above food levels has not been studied in pregnancy, breastfeeding or childhood, so food-level intake is the sensible ceiling there. Polyamines including spermidine support cell growth, and levels run high in some tumors, so anyone with an active cancer should raise supplemental use with their oncology team before starting. Food intake as part of a normal diet is a separate matter and is not the concern here.
